I wouldn’t ever have somebody come in and write the last draft of my novel or short story, and so right now I can’t imagine ever not editing one of my movies. Then you’re working with the music and a composer and taking away or adding sound, and for me, that’s all just another part of the storytelling. You’re trying to make their best stuff even better. “You’re still working with the actors - they’re not there, but you’re using different takes to put together a performance they never gave. Sayles added that editing his own movies is less about economics than maintaining authorial control in the way that writing novels accustomed him to. After transfer, stain the SDS-PAGE gel with Coommassie blueĪnother simple way to monitor the efficiency of protein transfer is to stain the gel with Coommassie after your transfer is complete. Remember, if you’ve opened your transfer cassette to “peek” at the ladder, this will introduce air bubbles to the transfer stack – so you probably want to use this technique to optimize transfer times when you’re not running important samples. If you use a ladder where each band is stained a different color, this can help you keep track of the transfer efficiency of differently sized proteins. Instead, you are aiming for a point at which most of the larger proteins have migrated to the membrane, and most of the smaller proteins are still on the membrane but not through it. Keep in mind that proteins of different sizes will migrate at different speeds: there is no magic moment at which all the proteins jump from the gel to the membrane. The rest of the proteins on your gel will of course be invisible, but you can easily see if the brightly colored ladder bands are still in the gel, or have transferred completely to the membrane. Use a pre-stained protein ladder to track transfer of proteins from a gel to a membrane. So how do you know when your transfer is complete? Here are a few good controls you can use to monitor transfer efficiency: Look for complete transfer of the molecular weight ladder Instead, you will need to optimize the transfer time and conditions for yourself, based on the equipment you use and the proteins you are working with. Transfer times are empirical and based on your own particular samples, which means that there is no easy way to determine how long you need to run the transfer in order to get complete transfer of all your proteins. Now it’s time for the all-important transfer step, that tricky point that will determine the quality of your Western blot. So, you’ve done your experiment, prepped your samples, and run your SDS-PAGE gel. 3 Again the paper has a lot of detail and is perhaps again summed up best by the authors: This systematic review was followed up by a direct study investigating blood type diet and its impact on cardiometabolic risk factors in 1,455 individuals. None of the studies showed an association between ABO blood type diets and health-related outcomes. 2 The paper itself is quite difficult to follow so I’ll just take this quote from the authors: 1 After screening 1,415 studies they identified 16 possible targets, which the further refined down to a single study of interest. In 2013, a group of researchers undertook just such a study into blood type diets. Scientists can perform a type of study known as a systematic review, where they pool the data from all relevant studies (not just those looking at the specific effect, but also other studies where appropriate data is recorded) and then analyse the effect. Which means a study with “sufficient power” is unlikely to be performed for a dietary intervention. Studies of this size are expensive and very time consuming and so are unlikely to be performed unless they are covering a major health risk. While it’s relatively straightforward to perform a study with 10 or 100 people, it becomes a completely different ball game when you reach 1,000 people or more. To investigate definitively, researchers need to make sure their study is sufficiently powered, which means they need lots of people to perform their comparison. Issues like this frequently occur in medical research when the underlying effect is multifactorial i.e. Whilst a lot of claims were made about the diet, clinical studies were lacking. If you’re planning to hang them on the tree, attach another loop of fishing line to use as a hanger. Just thread the two jingle bells onto the fishing line, tie a quick knot to keep the bells from separating, then tie them around the candy cane. I used fishing line to attach the jingle bells to the candy cane. You can put them in the fridge or freezer if it’s not cold enough outside, but I do recommend keeping them outside for at least a few minutes to let the melted plastic smell dissipate a bit before you bring them inside. I let them sit outside in the cold for about 10 minutes to cool completely. So if you make these at a warmer temperature, it will probably take less time for them to melt. Keep in mind that since I live in Canada, it was really cold outside when I made these with a temperature of 2✬ (35✯). I baked them for about 7 or 8 minutes outside until they had fused together. But it worked like a charm, and at least I didn’t have to worry about toxins in the house! You can melt them on your barbecue as well. I used 12 beads in each candy cane, alternating the colours as I went.Īnd again I got to look like a crazy person by putting our toaster oven outside on the porch to keep the plastic vapours out of the house. Yesterday after dinner, we had to make a family trip to Walmart to buy two of the three kids new winter boots (MAN, their feet grow fast!), and we were playing the kids favourite version of Santa Claus is Coming to Town in the van on the way to the store. Picture the tiniest flurry and my husband saying, “Debbie, it’s way too early for Christmas music!” and me replying, “What are you talking about!? It’s snowing!!!!!!” Even if it starts to snow in October, I’m excited for the excuse to play my favourite Christmas songs!!
